This practical guide for subsampling and preparing forensic samples for pollen analysis presents standard procedures for both the palynologist and nonspecialist technician.
Abstract
The primary forensic application of pollen analysis (palynology) is in providing evidence that will prove or disprove a link (type of pollen) between people and objects with places or with other people. This guide describes standard procedures for subsampling various types of materials, most of which are commonly collected for forensic analysis. The subsampling procedures are described for soil; clothing and other fabrics; footwear; twine and rope; air filters; firearms and tools; powders, crystals, and other granulated materials; plant material; and human and other animal material. Also included in this guide is a step-by-step method for concentrating pollen in the samples, based on the standard acetylation method. This includes potassium hydroxide digestion, sodium pyrophosphate treatment, hydrochloric acid treatment, hydrofluoric acid treatment, oxidation, acetolysis, staining, mounting, and the storage of prepared slides and pollen residues. 18 references
