This study examines the use of a loading dose of Divalproex (DVPX) in the potential treatment for cocaine dependence due to its possible enhancement of GABAergic activity, thereby decreasing cocaine use.

Abstract

This pilot study was designed to evaluate specifically safety and tolerability of the use of a loading dose of divalproex (DVPX) for the treatment of cocaine-dependant individuals. The study was an 8-week, open-label treatment using a loading dose (20mg/kg/day) of DVPX. Study subjects consisted of 17 males and females who met cocaine dependence criteria. During the study period, DVPX loading decreased the amount and frequency of cocaine craving and the percent of days using cocaine. Despite excluding major psychiatric illnesses, such as current affective, psychotic, and anxiety disorders significant improvement in psychological functioning was found at weeks 4 and 8. DVPX was seen as safe and well-tolerated in this population of cocaine users. Specific study limitations are discussed. References
