Abstract
This article discusses an optimized method to concentrate and clean up fluid extracts from the organic extraction of DNA from biological samples as an alternative to standard microdialysis filtration. The new method uses NucleoSpin XS silica columns to concentrate and clean the extracts. The method can be used both with standard organic extraction and dithiothreitol (DTT)-based differential extraction methods without making any modifications to the methods prior to concentration and cleanup. This article reports on the tests of the silica-binding method which uses a silica-membrane with a reduced effective membrane diameter. The reduced diameter membrane allows for low elution volumes making it attractive for use in low yield samples that are often encountered in forensic samples. The tests found that the silica-binding method could be used effectively as an alternative to the standard microdialysis filtration methods. Figures, table, and references
