The use of capillary array electrophoresis (CAE) as an approach to high-throughput DNA typing was examined in a research project that combined the rapid analysis times of capillary electrophoresis with the throughput multiplier of running many capillaries in parallel.
Abstract
Results revealed the usefulness of CAE for the high-throughput typing of short tandem repeat polymorphisms, both singly and in multiplex sets. The research also demonstrated the usefulness of CAE for high-throughput DNA sequence analysis. Findings also indicated that the use of energy transfer fluorescent labels improves both the sensitivity and the quality of analytical output, both for short tandem repeat typing and for sequence analysis. The researchers also constructed a second-generation CAE unit with four-color detection potential for both short tandem repeat typing and for sequence analysis. Additional research focused on other aspects of short tandem repeat typing, the detection of sample mixtures, and the potential of another genetic typing approach.
